AllCells LeukoPaks are leukapheresis products collected using the Spectra Optia® Apheresis System from healthy, consented donors following IRB-approved protocols. Depending on donor and customer needs, collections of apheresed cells are collected from a full procedure (2-2.5 blood volumes; ~120mL autologous plasma) or a half procedure (1-1.5 blood volumes; ~60mL autologous plasma). During the procedure, MNC rich cells are continuously collected into a sterile bag using ACD-A as an anticoagulant. Autologous plasma is then automatically collected directly into the cell pool following leukapheresis. As an option, apheresed cells can be provided without plasma to reduce processing time upon receipt. Apheresed total nucleated cell (TNC) products typically contain up to 50% T cells, 20% monocytes, 10% B cells, and 10% NK cells, 3% granulocytes, and 3% hematocrit.
Volume-Based: Our volume-based products meet the requirements for experiments where a minimum volume of apheresed cells is needed and where having a guaranteed number of cells is not a priority. After a sample is withdrawn for quality testing, a nominal volume of 290mL (Full Pak), 145ml (Half Pak) or 72mL (Quarter Pak) is transferred into a sterile transfer bag prior to shipping. These products are transferred into a sterile blood bag to contain a nominal, calculated volume of a collection, regardless of the cell count. The total nucleated cell count is determined using AO/PI from the collection and provided as information. Average full procedures produce ~17.5B TNCs with >90% of collections having more than 12.5B TNCs. These products are released based on the volume of apheresed material provided. The actual TNC concentration and cell total count will vary from collection to collection. Quarter and Half packs may be derived from either a fraction of a full or half apheresis collection.
Count-Based: When your experiment requires a minimum number of cells to be successful, our guaranteed cell quantity TNC products meet that need. Following collection, a total nucleated cell count is determined using AO/PI. A defined quantity of apheresed cells (2.5B, 5B or 10B cells, including a target overfill) is transferred into sterile containers. The total volume and concentration of the supplied cells will vary, but the number of cells you receive to begin your research is guaranteed*** to meet the label claim. On occasion (<10% of the time) the minimum number of cells is not achieved. In these rare cases, we will work with you to reschedule another collection to deliver the number of cells you expect.
